Spermidine is a polyamine that was first discovered in 1678. It may be best known for supporting autophagy, which can be thought of as a cell's natural recycling program. In addition to autophagy, spermidine plays important roles in responding to stress (as part of the polyamine stress response (PSR), and supporting cellular growth, DNA stability, brain health, and tissue growth and repair. Spermidine is found in many foods, including wheat germs, nuts, soybeans, mushrooms, and fermented products. Polyamine-rich foods—foods containing higher amounts of spermidine and other polyamines—have been linked to healthier aging in a range of organisms [1].

Spermidine can be obtained from the diet. It can also be made within the body. Despite these two ways of supplying it, the concentrations in some tissues (including the brain) of spermidine and other polyamines may decrease with increasing age [2,3]. Supplementing some additional spermidine, or eating more polyamine-rich foods, can act as a way to augment or refill polyamine pools that may decline during aging in various tissues. Higher intake of spermidine has been associated with healthier brain structure in older adults, as an example [4]. In human studies, spermidine has been supplied in amounts ranging from about ½ a mg to 3 mg [5–7].
